Why large dogs age faster: the pattern in the data

Growth speed and the IGF-1 link

The leading explanation is growth itself. A large-breed puppy performs a staggering feat, going from a couple of pounds to 100 or more in twelve months, which means its cells divide fast and often during that first year. The hormone behind that push is insulin-like growth factor 1, or IGF-1. A landmark 2007 study in Science traced small body size in dogs to a single IGF1 gene variant carried by nearly every small breed and almost no giant one, and that variant goes with much lower circulating IGF-1. Less growth hormone, a smaller dog, and a longer life.

High IGF-1 is a double-edged signal. It builds a big body quickly, but across many species it also raises the risk of age-related disease. Recent work has added a note of caution about cause and effect: it may be that large dogs die young because they carry more IGF-1, with their size partly a side effect of the same hormone rather than the direct killer. Its 2024 analysis, released as a preprint, found that larger breeds show faster epigenetic aging, a molecular clock read from chemical marks on DNA, matching the shorter lifespans seen in the field. Curiously, big dogs do not appear to age faster in the brain. In many big dogs, other problems end life before any dementia has a chance to appear.

Small terriers top the list near 12.7 years, large working breeds like the German Shepherd and Boxer sit around 10, and the size gradient is clear. The two flat-faced breeds break it. The Pug and the French Bulldog are small dogs that should live long, yet the French Bulldog lands last at 4.5 years.
